If someone in your family takes glucosamine for achy knees and also has mild cognitive impairment or Alzheimer’s disease, this is worth putting on the doctor’s question list.

Not the panic list. The question list.

A June 2026 paper in Nature Metabolism reported a new Alzheimer’s-related metabolism signal involving hyperglycosylation, a process tied to sugar-like molecular tagging in cells. In the same study, oral glucosamine moved cognitive outcomes in the wrong direction in Alzheimer’s-model mice. And in retrospective health-record data, glucosamine use was associated with worse outcomes among people already in the mild cognitive impairment, Alzheimer’s disease, or Alzheimer’s-related dementia spectrum.

That’s a serious enough signal to talk about. But it isn’t proof that glucosamine causes Alzheimer’s, and it doesn’t mean every person taking a joint supplement should abruptly stop it.

The more useful takeaway is narrower: disease stage matters. A supplement question that looks one way in generally healthy adults may look different after cognitive impairment or Alzheimer’s disease has already begun.
